Video Summary
Yaqiu Wang presented a new cell death pathway called <strong>mitotoxic pyrolysis</strong>, linking <strong>innate immune activation</strong> with <strong>metabolic starvation</strong>. In bone marrow-derived macrophages, combining <strong>Toll-like receptor stimulation</strong> with <strong>carbon-starvation media</strong> caused strong lytic cell death, while either condition alone did not. Known death pathways such as apoptosis, pyroptosis, necroptosis, and ferroptosis were ruled out. The key mechanism involved <strong>oxidative stress from mitochondria</strong>: mitochondria became trapped at the cell periphery, causing <strong>localized lipid peroxidation</strong> and membrane damage until the cell burst. <strong>M2 inhibitors</strong> reduced death by restoring mitochondrial mobility and actin-dependent membrane repair. In mice, combining <strong>fasting and LPS</strong> produced strong tumor-killing effects, suggesting therapeutic potential for cancer. Wang also noted ongoing work to identify the exact effector proteins and explore roles in infection.
